2 月 15 日,国际学术期刊 The Journal of Biological Chemistry 在线发表了中科院上海生科院(健康院)尹慧勇组的研究论文:“Identification of a Novel Series of Anti-inflammatory and Anti-oxidative Phospholipid Oxidation Products Containing Cyclopentenone Moiety in vitro and in vivo: Implication in Atherosclerosis”。该研究在低密度脂蛋白(low density lipoprotein, LDL)中发现一类新的磷脂氧化产物,通过细胞、动物和临床实验研究证明这类氧化产物具有抗炎抗氧化的作用,以及对动脉粥样硬化的保护作用。

心血管疾病是目前全球死亡率最高的疾病,动脉粥样硬化是其发病和死亡的主要病因。在动脉粥样硬化发生发展过程中,炎症和氧化应激发挥重要的作用。近年来在动脉粥样硬化斑块中发现多种磷脂氧化产物,并证实这些氧化产物通过与血管壁中的内皮细胞、巨噬细胞、平滑肌细胞等相互作用,参与动脉粥样硬化发展的每个阶段。但是,由于这些氧化产物具有高度的异质性,所以促进和抑制动脉粥样硬化的作用均有报道。

博士研究生陆剑虹等在尹慧勇研究员的指导下,利用脂质组学技术在氧化的低密度脂蛋白(LDL)和人动脉粥样硬化斑块中发现了一类新的磷脂氧化产物 deoxy-A2/J2-IsoP-PC,并且其水平在心血管病人血浆中是下降的。通过独立化学合成 15d-PGJ2-PC 作为代表分子研究这类新产物的生物学功能:研究发现 15d-PGJ2-PC 通过调控 NF-κB, PPARγ和 Nrf2 信号通路,诱导巨噬细胞抗炎抗氧化应答;在 LPS 诱导的系统性炎症小鼠模型中,15d-PGJ2-PC 减轻多个组织的炎症反应。此外,15d-PGJ2-PC 通过下调 ERK/SR-A 抑制巨噬细胞泡沫细胞的形成。

该研究工作得到了北大医学院郑乐民教授课题组、首都医科大学天坛医院、美国范德堡大学、加州大学圣地亚哥分校等合作者的大力支持,受到了国家自然基金、中科院和科技部等项目经费的支持。(科研处)
