流行病学研究中已经观察到咖啡摄入量与骨矿物质密度(BMD)之间的关联并不一致。此外,尚未有研究报道咖啡中生物活性成分与BMD之间的关系。近日,内分泌和代谢性疾病领域权威杂志Journal of Clinical Endocrinology & Metabolism上发表了一篇研究文章,该研究的目的是确定咖啡相关代谢物,并评估其与BMD之间的关系。

研究人员纳入了564名健康的社区居民。FFQ问卷中报告了咖啡消耗量。研究人员使用LC-MS平台对空腹血清样品进行非靶向代谢组学分析。通过DXA测定腰椎和股骨颈BMD。多变量线性回归和稳健回归用于关联分析。

在Bonferroni校正后的多重分析中,血清12种代谢产物与咖啡摄入呈正相关(p<4.87×10-5),其中奎宁酸、3-羟基吡啶硫酸盐和葫芦巴碱显示相关性最强。在这些代谢产物中,先前已确定11种已知代谢产物与咖啡摄入有关,其中6种与咖啡因代谢有关。习惯性的咖啡摄入量与腰椎和股骨颈的BMD呈显著正相关。5-乙酰氨基-6-甲酰基氨基-3-甲基尿嘧啶(AFMU)(β=0.012,SE=0.005,p=0.013)与腰椎BMD显著相关,而3-羟基马尿酸盐(β=0.007,SE=0.003,p=0.027)和曲古碱(β=0.007,SE=0.004,p=0.043)与股骨颈BMD显著相关。

由此可见,12种代谢产物与咖啡摄入量显著相关,包括6种咖啡因代谢产物。它们中的三个(AFMU、3-羟基马尿酸盐和葫芦巴碱)进一步与BMD相关。这些代谢物可能是咖啡摄入的潜在生物标志物,并影响骨骼健康。
