J Dent Res：蛋白质精氨酸甲基转移酶PRMT1对于腭板形成是必需的

腭裂是最常见的先天缺陷。目前，只有30％的病例确定了遗传原因，而大多数病因仍有待发现。我们鉴定了一种新的腭发育调节因子蛋白质精氨酸甲基转移酶1（PRMT1），并证明神经嵴细胞中PRMT1功能的破坏导致完全腭裂和颅面畸形。PRMT1是蛋白质精氨酸甲基转移酶中最高表达的，该酶是负责组蛋白和非组蛋白上精氨酸基序甲基化的酶。PRMT1调节信号转导和转录活性，影响颅面发育中至关重要的多种信号通路，如BMP，

我们鉴定了一种新的腭发育调节因子蛋白质精氨酸甲基转移酶1（PRMT1），并证明神经嵴细胞中PRMT1功能的破坏导致完全腭裂和颅面畸形。PRMT1是蛋白质精氨酸甲基转移酶中最高表达的，该酶是负责组蛋白和非组蛋白上精氨酸基序甲基化的酶。PRMT1调节信号转导和转录活性，影响颅面发育中至关重要的多种信号通路，如BMP，TGFβ和WNT通路。我们证明了Wnt1-Cre; Prmt1 fl/fl小鼠显示腭间充质细胞增殖减少和腭架未能到达中线。在信号通路中的进一步分析显示突变小鼠中Prmt1的丧失降低了BMP信号传导活化并减少了H4R3me2a标记的沉积。
