Blood:基于代谢状态的癌症治疗方法——Aldh3a2抑制

恶性细胞(相对于正常细胞)的体内代谢依赖关系可以通过体外筛选来确定。 Aldh3a2与GPX4抑制是综合致死的,提供了一种仅基于代谢状态的联合治疗方法。

癌症的代谢改变代表了致癌突变的聚合效应。研究人员假设,在模拟骨髓微环境的体外系统中,通过对正常小鼠造血细胞和恶性造血细胞进行代谢受限的遗传筛选,可以明确急性髓系白血病(AML)的独特弱点。

白血病细胞,但不是正常的骨髓细胞,依赖于醛脱氢酶3a2 (Aldh3a2);Aldh3a2能氧化长链脂肪族醛,以防止细胞氧化损伤。醛类化合物是癌症中氧化磷酸化和核苷酸合成增加的副产物,由脂质过氧化物产生,而脂质过氧化物是细胞死亡的非caspase依赖性形式,即铁中毒。

白血病细胞对Aldh3a2的依赖性可见于多种小鼠和人类髓系白血病。Aldh3a2抑制与谷胱甘肽过氧化物酶-4 (GPX4)抑制联合起来对AML细胞来说是致命的;GPX4抑制是一种已知的对AML细胞影响最小的铁功能触发。

总而言之,抑制Aldh3a2为AML提供了一个治疗机会,以独特的合成致死性机制靶向独特的代谢状态的恶性细胞。
