J Endod:牙髓局部应用抗生素的抗菌效能及变色潜力

牙髓局部应用抗生素的最佳浓度还未可知。这篇研究的目的是为了检测甲硝锉、环丙沙星、米诺环素、力百汀(GlaxoSmithKline, Research Triangle Park, NC)和替吉环素对抗牙髓常见致病菌的最小杀菌浓度(MBCs)和最小抑菌浓度(MICs),并通过缓释水凝胶支架体外评估三重抗生素糊剂(TAP)、力百汀和替吉环素在不同浓度下的抗菌效能和变色影响。

研究使用Epsilometer测试法(Etest; bioMérieux USA, St Louis, MO), 所选抗生素的MICs和MBCs来拮抗具核梭杆菌、牙龈卟啉单胞菌、中链球菌、粪肠球菌。将这些菌属的生物膜置于厌氧环境下拔除的单根牙齿内3周。随后,使用富含1或0.1 mg/mL的TAP、力百汀和替吉环素的可降解水凝胶支架或单纯1 g/mL的TAP充填根管7天。在充填修复后的1, 2和3周通过光谱广度测量法评估牙冠变色情况。

MIC/MBC数据显示,与其他抗生素相比较,米诺环素、力百汀和替吉环素展现出明显的药效(P < .05)。所有实验组的log10菌落形成单位之间均存在显著性差异(P < .05)。在1 g/mL TAP作用下没有细菌生长但会导致最大程度的变色。与导致最小变色的药物浓度0.1 mg/mL (P < .05)相比,1 mg/mL的TAP、力百汀或替吉环素与水凝胶混合物可以显著减小细菌生长和阳性样本数量。

结论:水凝胶中的TAP、力百汀或替吉环素在1 mg/mL的浓度时可以显著减弱细菌生长以及最低程度的导致变色。
