我们是怎么知道我们身在何方的?我们怎么找到从一个地方到另一个地方的路线的?我们是怎么把这些信息储存下来,让我们下次重返故地的时候能立刻找到路?

2014年诺贝尔生理学或医学奖的得奖者发现了大脑里的“定位系统”,一台内置的GPS,使得我们能在空间中定位自己身在何处,这表明这种高级认知功能,如空间认知和位置认知也有细胞级别的基础。

1971 年,约翰·奥基夫发现了这个定位系统的第一个成分。奥基夫发现大脑内侧颞叶-海马的神经元动作电位(快活动)与膜电位波动(慢活动,脑电EEG的基础)之间有规律移动的锁相关系(相位递进现象),为EEG参与神经信息加工提供了证据。过去认为只有动作电位参与。他发现,大脑海马体里有一种神经细胞,每当大鼠身处屋子的某个特定地点的时候,这种细胞总是会被激活。 其它神经细胞则在大鼠身处其它地方的时候被激活。奥基夫的结论是,这些“位置细胞”(place cells)组成了屋子的地图。

三十多 年后,迈-布里特·莫泽和爱德华·莫泽发现了大脑定位系统的另一个关键成分。他们发现了另一种神经细胞,命名为“网格细胞”(grid cells),它们组成了一个坐标系,允许生物进行精确的定位和寻路。他们的后续研究表明,地点细胞和网格细胞一起使得定位和导航成为可能。

约翰·奥基夫,迈-布里特·莫泽和爱德华·莫泽的研究回答了困扰哲学家和科学家数百年的问题——大脑如何给周围的空间创造地图,我们如何在复杂的环境中寻找路线。

我们如何感知我们所在的环境?


人存于世,位置感和导航能力是不可或缺的。位置感使我们能够感知自己在环境中所处的位置。在导航时,我们的位置感会与基于运动和对先前位置认知所形成的距离感相互联系起来。

关于地点和导航的问题困扰了哲学家和科学家许久许久。200多年前,德国哲学家康德认为一些精神能力是独立于经验的先天知识。他认为空间概念是意识中既有的原则,人们会通过,也必须通过这些原则感知世界。

到 20世纪中叶,行为科学的出现使得这些问题得以通过实验手段进行解答。当爱德华·托尔曼(Edward Tolman)观察迷宫中大鼠的运动时,他发现它们能够学习如何导航,并提出它们脑中形成了一副“认知地图”,使它们找到自己要去的路。但问题并未完全解 决:这个“地图”在大脑中的表征是什么?

约翰-奥基夫以及空间位置

在20世纪60年代晚期,约翰-奥基夫对于大脑如何控制行为和决策这一问题十分着迷,并常试图用神经生理学的方式来解决这一问题。当他记录在屋内自由跑动 的大鼠的大脑海马体内单个神经细胞的信号时,奥基夫发现,当大鼠经过特定位置时,某些神经细胞会被激活。他发现这些“位置细胞”不仅仅接受视觉信号输入, 而且还会在脑中绘制周围环境的地图。奥基夫总结道,通过在不同环境中被激活的不同的位置细胞,海马体能生成很多地图。因此,关于环境的记忆能以位置细胞活 性的特定组合形式被存在海马体中。

迈-布里特-莫泽和爱德华-莫泽找到了定位系统

迈 -布里特-莫泽和爱德华-莫泽在绘制移动中的大鼠的海马体连接时,在附近的内嗅皮层中发现了一种让人惊异的活动模式。当小鼠通过六角网格中的某些位置时, 内嗅皮层中的某些固定的细胞会被激活。每个细胞都对应着某个特定的空间格局,这些“网格细胞”共同建立出一个可以进行空间导航的坐标系统。它们和内嗅皮层 中其他负责辨识头部方向和房间边界的细胞一起,与海马体中的位置细胞共同组成了神经回路。这个回路系统在大脑中建立了一套综合定位系统,一个内置的 GPS。

人类大脑里的“地图”

根 据最近的脑成像技术调查,以及对接受神经外科手术患者的研究都显示,位置细胞同样存在于人体中。在早期阶段阿尔兹海默氏疾病的早期阶段,患者的海马体和内 嗅皮质经常会受到影响,以致这些患者经常无法辨别周边环境并且迷路。了解大脑的位置系统或许可以因此帮助我们了解这种疾病如何对患者的空间记忆丧失造成影 响。

这一对大脑位置系统的发现代表了我们进一步认识大脑特化细胞如何协同合作,并执行更高水平的认知功能。它为我们理解认知过程,比如记忆、思维与计划开辟了新的途径。

三位获奖者介绍:

John O'Keefe
,1939年出生于美国纽约,拥有美国和英国双重国籍。1967年他从加拿大麦吉尔大学获得生理心理学博士学位。之后,他进入伦敦大学学院做博后。1987年被UCL任命为认知神经学教授。目前仍任职于UCL。

May-Britt Moser
,1963年出生于挪威福斯纳瓦格,挪威公民。曾在挪威奥斯陆大学学习心理学。1995年获得神经生理学博士学位。2000年被挪威科技大学任命为神经学教授。目前为挪威特隆赫姆神经计算中心主任。

Edvard I. Moser
,1962年出生于挪威奥勒松,挪威公民。1995年从挪威奥斯陆大学获得神经生理学博士学位。Edvard I. Mosel和May-Britt Moser为夫妻。二人曾共同在爱丁堡大学做博后。1996年他加入挪威科技大学,1998年成为教授。目前为挪威特隆赫姆Kavli系统神经学研究所主任。
