Circulation: P2X7敲除抑制斑块内炎症小体活性并改善小鼠动脉粥样硬化

单独使用LPS或ATP刺激不能激活离体巨噬细胞中凋亡蛋白酶1。然而,用LPS和ATP序贯刺激导致P2X7-补足的巨噬细胞中凋亡蛋白酶1和IL-1的活化。相比之下,P2X7敲除的巨噬细胞在连续刺激后没有凋亡蛋白酶1的活化,同时表达基础量的IL-1β。P2X7受体在鼠动脉粥样硬化病变中表达较高,特别是在斑块内巨噬细胞。相比于P2X7补足的小鼠(0.162cm2±0.023(n = 9)),给予P2X7敲除小鼠16周高胆固醇饮食后(0.084cm2±0.01(n = 11))表现出更小的动脉粥样硬化病变(p = 0.004),斑块内巨噬细胞数量也减少。与体外研究结果相一致,P2X7 - / - 小鼠中凋亡蛋白酶1的活性被消除。此外,活体显微镜显示P2X7敲除小鼠中白细胞滚动和粘附减少。最后,我们发现人动脉粥样硬化斑块中P2X7表达增加,表明我们在小鼠中的发现与人类疾病一致。

P2X7敲除通过抑制斑块内炎症细胞活化逆转炎症,减少实验动物动脉粥样硬化。因此,P2X7是对抗动脉粥样硬化的潜在新靶点。
