本研究的目的是开发一种临床上侵入性较小的方法,分离和扩增人SG干细胞,获得无细胞提取物,用于治疗IR损伤的SG。实验中,对人的唇腺进行活检,并通过体外培养扩增唇腺干细胞(LSC)。通过3次冻融循环和17,000g离心后获得LSC提取物(LSCE)。将LSCE静脉注射到13-Gy IR处理SGs的小鼠中。阳性(non-IR)和阴性(IR)对照小鼠均注射了生理盐水(对照)。三片唇腺(0.1克重)最终可以获得100-200万个细胞。LSC的倍增时间为18.8小时;可以分化为骨细胞,脂肪细胞和软骨细胞;间充质干细胞标记阳性。

结果发现,在LSCE中可以检测到血管生成因子(FGF-1,FGF-2,KGF,血管生成素,uPA,VEGF)和抗血管生成因子(PAI-1,TIMP-1,TSP-1,CD26)。此外,一些血管生成因子(PEDF,PTX3,VEGF)具有神经营养功能。在IR后的8周和12周,用LSCE处理的小鼠的唾液流速比经盐水处理的小鼠高50%至60%。唾液滞后时间测量的实验也证实LSCE可以恢复SG功能。腮腺和下颌下腺的组织学分析表明,在non-IR和LSCE处理的小鼠中,腺泡细胞、血管和副交感神经的数量和细胞增殖率相当,尽管在盐水处理的小鼠中明显降低。

体外培养的方法可以从小块的腺体中收获大量LSC。LSCE具有减轻IR损伤的SG的临床潜力。
