来自南非斯坦堡大学(SU)的ResiaPretorius教授和曼彻斯特大学的道格拉斯·凯尔教授进行了一系列研究，大大改变了科学家们关于细菌对许多疾病(的影响的思考方式，包括阿尔兹海默、帕金森病，败血症，类风湿关节炎和最近的2型糖尿病(T2D)。

以前，Pretorius和Kell已经确定，这些慢性炎性疾病也具有微生物来源。凯尔说：“如果细菌是活跃的或复制的，就像传染病一样，我们将会了解这一切。“但微生物不是复制的，它们主要是休眠的。”

因为它们的休眠性质意味着它们在标准的微生物测试条件下能够隐藏，所以以前认为细菌不存在于人的血液中。然而，高水平的铁(典型的炎性疾病)可以有效地使这些细菌恢复生命。以前的研究表明，在这些条件下，细菌开始复制和分泌脂多糖(LPS)，导致炎症增加。

这些慢性疾病有一个共同点是炎症水平不断提高。Pretorius和Kell已经证实了异常的淀粉样变血液凝固与细菌细胞壁组分如LPS和脂磷壁酸(LTA)相关并可能由实验诱导。

这些凝血病(不良血液凝固)也是典型的炎性疾病，研究人员长期以来表明它们导致淀粉样蛋白形成，其中血液凝固蛋白(称为纤维蛋白原)在结构上从α-螺旋形变形为平板状的b片状结构，可能导致细胞死亡和神经变性。

结果，患病个体血凝块的纤维蛋白与健康个体明显不同。这可以在显微镜下进行可视化。“在正常的血块中，这些纤维看起来像一碗意大利面条”，Pretorius解释说。“但是在患病个体中，他们的血块看起来很融合和凝聚纤维，也可以观察到在淀粉样蛋白存在下发出荧光的特殊污渍。”

研究人员发现，这种改变的凝块结构存在于研究的所有炎症状况中，现在包括2型糖尿病。但这种异常凝块形成，细菌，LPS和TLA之间的联系是什么?还有哪些分子可以“擦拭”LPS或LTA，并且可能会在具有炎性疾病的人的血液中循环?

在2017年的研究中，Pretorius和Kell，以及来自比勒陀利亚大学的MSc学生SthembileMbotwe女士研究了LPS-结合蛋白(LBP)的作用。他们将LBP添加到T2D患者的血液中(以及加入LPS后的健康血液)。以前他们已经表明LPS加入到健康的血液中会导致凝块形成异常，而LBP也可以逆转。在本试验中，他们表明，LBP也可逆转T2D血液中不良凝块结构。该过程通过扫描电子显微镜和超分辨率共聚焦显微镜证实。结论是明确的：细菌LPS是T2D发展和维持及其致残后遗症的重要参与者。

“在发炎的情况下，大量的LPS可能会阻止LBP正常工作，”Pretorius解释说。

研究人员总结说：“我们现在有相当多的证据，其中大部分是新的，与目前攻击T2D的策略相比，认识到它涉及休眠的微生物，为慢性炎症过程和凝血病，提供了新的治疗机会。
