免疫检查点抑制疗法为肿瘤治疗带来了革命。但是只有不到一半的接受治疗的患者出现持续缓解。由于缺少对启动抗肿瘤免疫反应的细胞特征的理解，如何改善治疗效果也一直并不明确。CLIN CANCER RES近期发表了一篇文章研究这一问题。

免疫检查点抑制疗法为肿瘤治疗带来了革命。但是只有不到一半的接受治疗的患者出现持续缓解。由于缺少对启动抗肿瘤免疫反应的细胞特征的理解，如何改善治疗效果也一直并不明确。CLIN CANCER RES近期发表了一篇文章研究这一问题。

作者对未接受过免疫治疗的黑色素瘤患者的肿瘤标本以及抗PD-1治疗期间连续性活检标本进行流式检测和免疫荧光染色。研究结果表明，CD69+CD103+肿瘤定植CD8+T细胞数目增加与未接受过免疫治疗的黑色素瘤患者黑色素瘤特异性生存改善有关。局部IL15表达水平与这些肿瘤定植T细胞数目有显着的相关性。包括PD-1和LAG3在内的几种免疫检查点表达增加，在抗PD-1免疫治疗早期这些细胞显着扩增。

文章最后认为，肿瘤定植CD8+T细胞数量比转移性黑色素瘤总CD8+T细胞数目预后作用更明显。此外，这些T细胞可能触发抗PD-1和抗LAG3治疗反应。在治疗前绘制这些细胞的免疫谱可以为免疫检查点抑制疗法提供更多信息。
