Blood:B细胞正常发育需要生理性沉默miR125b的表达

多种microRNAs调节异常可影响造血过程中的关键发育结点,以致影响细胞功能,最终导致自身免疫性疾病或癌症。近期,Guideng Li等人发现miR-125b在骨髓多能祖细胞和髓系细胞中表达,但在B细胞系中不表达,而且在B细胞中编码miR125b的基因缺乏转录激活标志物。为了解B细胞中生理性沉默miR-125b的生物学意义,研究人员在B细胞系中诱导其表达发现,miR-125b表达异常会损伤未成熟

多种microRNAs调节异常可影响造血过程中的关键发育结点,以致影响细胞功能,最终导致自身免疫性疾病或癌症。

近期,Guideng Li等人发现miR-125b在骨髓多能祖细胞和髓系细胞中表达,但在B细胞系中不表达,而且在B细胞中编码miR125b的基因缺乏转录激活标志物。

为了解B细胞中生理性沉默miR-125b的生物学意义,研究人员在B细胞系中诱导其表达发现,miR-125b表达异常会损伤未成熟B细胞从骨髓进入外周血的过程。该损伤主要是通过抑制鞘氨醇-1-磷酸受体1(S1PR1)来介导。增强S1PR1的表达或用CRISPR/Cas9介导编辑S1PR1 3'UTR端miR-125b靶向位点,从而挽救miR-125b介导的B细胞外迁缺陷。

除了会影响B细胞外迁,miR-125b失调先是降低前体B细胞输出,随后可诱导小鼠发生前体B细胞淋巴瘤/白血病。此外,研究人员在miR-125b诱导的B细胞癌中发现了IRF4基因缺失,但其在致癌性miR-125b诱导的B细胞转化中的作用尚不清楚。

研究人员又在IRF4纯合突变小鼠中发现miR-125b诱导的B细胞白血病进展加快,进一步证明了miR-125b和IRF4在癌症诱导中存在相互作用。

因此,Guideng Li等人认为B细胞正常发育需要生理性沉默miR-125b,而且miR-125b的生理性沉默也可作为抑制肿瘤发生的一种机制。
