Circulation:硒蛋白P可促进肺动脉高压进展

肺动脉平滑肌细胞(PASMCs)过度增殖和凋亡耐受是肺动脉高压(PAH)的关键机制。尽管现已有多种联合疗法,但因缺乏针对PASMCs的诊断生物标志物和抗增殖疗法,有很多患者恶化成重度肺动脉高压(PH)。现有研究人员采用微阵列分析来鉴定PAH的新的治疗靶点。研究人员以PAH患者的肺和血清样本、培养的PAH-PASMCs和高通量筛查探究机制和新的治疗药物;采用基因编辑鼠(包括PASMC特异性硒蛋白P[

肺动脉平滑肌细胞(PASMCs)过度增殖和凋亡耐受是肺动脉高压(PAH)的关键机制。尽管现已有多种联合疗法,但因缺乏针对PASMCs的诊断生物标志物和抗增殖疗法,有很多患者恶化成重度肺动脉高压(PH)。

现有研究人员采用微阵列分析来鉴定PAH的新的治疗靶点。研究人员以PAH患者的肺和血清样本、培养的PAH-PASMCs和高通量筛查探究机制和新的治疗药物;采用基因编辑鼠(包括PASMC特异性硒蛋白P[SeP]敲除小鼠和PH模型大鼠)模型来研究SeP的功能以及多种化合物对PAH/PH的治疗潜能。

研究发现PAH-PASMCs中的SeP比对照PASMCs中的SeP高32倍。SeP是广泛表达于细胞外的蛋白质,维持细胞代谢。在PAH患者的增厚的肺动脉中层中,SeP的免疫活性增强。此外,PH患者的血清SeP水平也升高,高SeP血清水平提示预后不良。持续性缺氧处理的SeP敲除小鼠(SeP-/-)表现为右心室收缩压显降著低,右心室肥厚和肺动脉重构。与此相反,全身过表达SeP的小鼠表现为低氧诱导型PH加重。而且,PASMC特异性SeP-/-小鼠表现为低氧诱导性PH减轻,而肝特异性SeP敲除或肝特异性过表达SeP小鼠均与对照组无明显差异。总体而言,肺内SeP蛋白水平与PH的进展相关。机制研究表明SeP可通过增加氧化应激和线粒体功能异常从而促进PASMC增殖和凋亡抵抗。此外,对3336种化合物进行高通量筛选发现血根碱(sanguinarine),一种具有抗增殖效应的生物碱,可降低SeP的表达、减少PASMCs增殖,缓解小鼠和大鼠的PH。

综上所述,SeP可促进PH进展,提示其或可作为肺动脉高压的新型生物标志物和治疗靶点。
