Sci Rep:支持1-碳循环的代谢增强子能够影响精子功能

最近,有研究人员将人类、牛和海鞘类精子在含有支持1-CC物质(维他命B6、甲基钴胺素、5-四氢叶酸甲酯、双甘氨酸锌和乙酰基半胱氨酸)(TRT)的培养基中孵育,并且与只含有乙酰基半胱氨酸(NAC)的培养基组进行比较。在对照组(CNTRL),精子只用培养基进行孵育。研究发现,90和180分钟的孵育后,在TRT和NAC中,线粒体膜电位(ΔΨM)要比CNTRL显著更高(P<0.01)。在H2DCFDA评估中,ROS在不同的种类中具有差别,而在2-OH乙啡啶中,ROS在牛TRT组中显著减少。另外,在不同的处理中,细胞内pH并没有显著的变化。在海鞘类精子中,NAC的补充能够减少细胞外pH,从而引起细胞内pH的降低。患有碳酸氢钠的缓冲液能够逆转NAC补充带来的益处。

最后,研究人员指出,在哺乳动物精子中,1-CC和NAC单独处理均能够改善代谢、ΔΨM和ROS产生,首次阐释了体外试验中,这些化合物对精子的功能具有直接的影响。
