结节病是一种不可预测的肉芽肿性疾病,其中部分非洲裔美国人(AAs)表现为侵袭性结节病。细胞对各种应激物产生反应释放出的线粒体DNA(mtDNA)有助于组织重塑和炎症。虽然细胞外mtDNA已成为多种疾病的生物标志物,但其与结节病的相关性仍然未知。近日,呼吸疾病领域权威杂志Eur Respir J上发表了一篇研究文章,研究人员的目的是确定细胞外线粒体DNA与结节病的临床特征之间的关联。

研究人员使用qPCR测量来自Yale Lung Repository的健康对照者和结节病受试者血浆样品和支气管肺泡(BAL)样本中人MT-ATP6基因,以衡量细胞外mtDNA浓度;并探究MT-ATP6浓度与Scadding阶段,肺外疾病和人口统计学指标之间的关联。在Alpha-1抗胰蛋白酶缺乏症和结节病队列的基因组研究中得到验证。

相对于对照者,结节病受试者BAL和血浆MT-ATP6浓度剧烈升高,特别是在患有肺外疾病的受试者的血浆中。相对于高加索人,AAs在BAL和血浆中显示出过量的MT-ATP6浓度,后者与显著较高的肺外疾病几率相关。 

由此可见,结节病中细胞外线粒体DNA的富集与肺外疾病和AA血统有关。进一步研究这些临床发现的机制可能会产生新的病理生理学和治疗理论。
