韩春雨面对质疑声称:实验没问题 已重复两次

国内外大量实验室为何无法重复其实验?“细胞污染”是韩春雨向同行和媒体常说的一个原因。  “别人可能会有疑问,怎么这么多人手里的细胞都是有问题的?我也从各种渠道购买,或者也从别人手里要了实验材料,把原来整个实验捋一遍,做一遍,确实发现,他们说做不出来确实是有原因的,我现在基本上确认其他实验室不能重复出来实验的一个很重要的原因是细胞污染,但我还要通过更严谨的实验去把这些东西找出来。”韩春雨解释道。

“别人可能会有疑问,怎么这么多人手里的细胞都是有问题的?我也从各种渠道购买,或者也从别人手里要了实验材料,把原来整个实验捋一遍,做一遍,确实发现,他们说做不出来确实是有原因的,我现在基本上确认其他实验室不能重复出来实验的一个很重要的原因是细胞污染,但我还要通过更严谨的实验去把这些东西找出来。”韩春雨解释道。

“细胞污染是一个挺严重的事情,它曾经对世界上很多科学家造成困扰,Nature有过专门的报道。”韩春雨强调说,“我做这个事情就是对12个实名科学家的回应。”

目前,韩春雨集中精力“在分析判断别人的(实验用)细胞和我手里的细胞”,“我希望找出确凿的污染源,这样的话别人也能做出来”。

据韩春雨实验室的一位工作人员说,韩老师每天都在实验室泡着,晚上很晚才回家。在与《知识分子》通电话的当晚,一直工作到次日凌晨三点。

韩春雨告诉《知识分子》,自己目前最主要的时间都花在做实验上,“但是做实验的时间也不多”,“我整个八月份几乎没法在实验室干活,总有电话,会有记者堵门”。“我需要时间,需要一个安静的环境”。

当《知识分子》再次问及在其成功重复的两次实验中,是如何处理细胞污染问题时,韩春雨表示,自己在发《自然?生物技术》论文之前也遇到过类似的细胞污染的问题,但拒绝透露自己解决问题的“秘诀”,“我只能说将来我会以比较严谨的科学报告的方式去(回应)”。

究竟谁重复了实验?

2016年8月,经历国内外多家实验室无法重复实验的质疑高峰后,韩春雨接受了自然新闻亚太通讯员DavidCyranoski的采访,并提供了5位据称可以重复实验的中国科研人员名单。

最终有三位匿名的中国科研人员出现在自然新闻的报道中。其中一位表示在好几个细胞系检测了NgAgo系统,而且结果显示NgAgo能够在预期的位点诱导遗传突变,但NgAgo系统的效率并没有比CRISPR-Cas9高,可能还要后续调整改进。另有两名科学家称有了一些初步的试验结果显示NgAgo是有效的,但是仍然需要进一步测序去确认。

9月6日,《成都商报》刊发了对韩春雨的专访,韩春雨称“《自然》杂志已经证实实验可重复,但还有不确定因素影响实验结果”。随后,DavidCyranoski本人表示自然新闻的报道不能作为韩春雨实验可重复的证据。

究竟是谁已经重复了实验,韩春雨对此一直讳莫如深,不愿多谈,只是声称河北科技大学也“明确知道哪几家(实验室)做出来了,和实验室都有接触”。

当《知识分子》追问:能够重复的实验室是如何解决细胞污染问题的?韩春雨欲言又止,表示这一问题“涉及到人家的机密”,并补充道:“都是在某些特别干净的细胞里做出来的”。

在接受《知识分子》访问时,韩春雨拒绝公开究竟谁重复了实验,理由是“因为前一段媒体报道,做出来就会被骚扰一通这个事很不好”。

事实上,由于外界普遍不知哪个实验室重复韩春雨实验结果成功,这样的骚扰是否发生无法求证。韩春雨坚持认为公开已经重复实验的实验室并不是好办法,他向《知识分子》描述了心目中解决问题的最好办法——自己做实验“解决掉一些问题,或者提出一个办法”,再找另外的第三方实验室重复实验,既保全目前“已经重复实验”的合作伙伴,又解决大家不能重复存在的问题。

韩春雨说,这一过程“大概需要20天时间左右”,“20天的时间可以发生很多事情”,但他不愿详谈具体会发生什么。

浙大教授:韩春雨有责任澄清学术质疑

我是王立铭,浙江大学生命科学研究院教授、研究员、博士生导师,一名普通的生命科学研究者。在这里,我谨代表我自己,简单总结一下我们实验室和合作者测试检验这一基于NgAgo的全新基因编辑技术的过程。
