材料与方法:实验对象为6周龄无菌小鼠,每周两次口服给予致病菌牙龈卟啉单胞菌或口腔共生唾液乳杆菌,持续5周。对照小鼠仅给予赋形剂。组织学评估牙槽骨吸收情况。使用实时聚合酶链反应分析牙龈组织,肝脏,小肠和大肠中各种基因的表达谱。分析血清的抗体,内毒素和白细胞介素(IL)-6水平。还分析了来自肠系膜淋巴结和脾的细胞的培养上清液的抗体水平。通过流式细胞术分析来自肠系膜淋巴结和脾的细胞中T-辅助细胞17和Treg的比例。通过酶联免疫吸附测定分析细胞培养上清液中IL-6和IL-17的水平。

结果:牙龈卟啉单胞菌给药不会诱导牙槽骨吸收。尽管牙龈卟啉单胞菌在无菌小鼠中引发全身抗体反应,但与特定无病原体小鼠不同,牙龈卟啉单胞菌不会在牙龈,肝脏和肠组织中诱导炎症反应,或改变T辅助细胞17和Treg的比例。然而,给予牙龈卟啉单胞菌小鼠的肠系膜淋巴结的细胞中,IL-6和IL-17显着升高并且趋于升高。有趣的是,在牙龈卟啉单胞菌处理的小鼠中,IL-10和紧密连接蛋白在牙龈和肠中的表达均显着上调。唾液乳杆菌与牙龈卟啉单胞菌几乎引起相似的效果。

当给无菌小鼠施用时,口腔致病菌牙龈卟啉单胞菌不诱导任何可检测的致病性变化或任何主要宿主反应。牙龈卟啉单胞菌可能存在肠道介导的全身作用的间接机制。
