来自在莫斯科的nasonova风湿病研究所的Elena V. Tchetina博士和他的同事检测了26个对照受试者和40例RA患者以确定基线基因在整个外周血中表达的潜力,RA患者均接受改变病情抗风湿药物MTX治疗,以预测其对MTX治疗的应答。

研究人员发现,在研究结束时,随着疾病活动度的下降,关节缩窄分数(JSN)显着升高。UNC-51-样激酶1(ULK1)和基质金属蛋白酶(MMP-9)的基因表达和血清中C反应蛋白的水平呈正相关。MMP-9基因表达还和28个关节的疾病活动度评分(DAS28)以及关节肿胀技术相关。随访结束时,基线肿瘤坏死因子-α的表达和JSN的表达成正相关,而P12,半胱天冬酶3以及Runt相关转录因子2(RUNX)的表达和DAS28的变化相关,说明这些基因的表达和RA患者病情改变相关,即对治疗药物的应答相关。

该研究的作者说:“我们的研究提示,MMP-9和ULK1基因可能和RA的疾病活动度相关。另外,在基线水平,RUNX2、p21和半胱天冬酶3基因在外周血的表达可能预示着该患者对MTX治疗的反应较好。”
