CLIN CANCER RES：SL-101治疗急性髓系白血病的抗白血病效果及其机制

诱导治疗后白血病干细胞（LSC）的持续存在对微小残留病灶以及急性髓系白血病（AML）复发有促进作用。CLIN CANCER RES近期发表了一篇文章，研究CD34+CD123+LSC的临床作用以及靶向CD123+LSC抗体共舞SL101的抗白血病效果。

诱导治疗后白血病干细胞（LSC）的持续存在对微小残留病灶以及急性髓系白血病（AML）复发有促进作用。CLIN CANCER RES近期发表了一篇文章，研究CD34+CD123+LSC的临床作用以及靶向CD123+LSC抗体共舞SL101的抗白血病效果。

作者回顾性分析了86例新确诊的AML患者，发现高比例CD34+CD123+LSC与持续微小病灶残留有关，并且可以预测低遗传风险的患者较差的无复发生存情况。利用流式细胞学，作者探索了使用抗体共轭物SL-101靶向CD34+CD38-CD123+细胞的潜在治疗效果。通过AML细胞系CD123抗原表达水平决定SL-101的抗白血病能力。细胞集落形成实验表明SL-101有力地并选择性地抑制白血病干细胞，同时不会影响其他正常的成分。进一步的研究表明SL-101的细胞毒性活性机制包括抗体快速有效地内化，蛋白质合成的持续抑制，诱导凋亡以及阻断IL-3诱导的p-STAT5以及p-AKT信号通路。在NSG鼠异体移植瘤模型中，使用SL-101预处理过的LSC在体内的再生能力显着受损。

文章最后认为，该研究明确了SL-101靶向AML治疗的机制，为SL-101进一步的临床应用研究以及其他靶向CD123治疗AML治疗策略的临床应用提供了依据。
