J Endod:脂多糖通过诱导自噬下调根尖牙乳头来源干细胞的成骨/成牙本质分化

牙髓牙本质的组织再生过程中干细胞牙本质发生的潜力可能会受到微生物的影响。这篇研究的目的是通过体外评估根尖牙乳头来源的干细胞(SCAP)在施加细菌脂多糖(LPS)刺激时的细胞活性以及成骨/成牙本质分化潜力,并探讨其分子机制。

牙髓牙本质的组织再生过程中干细胞牙本质发生的潜力可能会受到微生物的影响。这篇研究的目的是通过体外评估根尖牙乳头来源的干细胞(SCAP)在施加细菌脂多糖(LPS)刺激时的细胞活性以及成骨/成牙本质分化潜力,并探讨其分子机制。研究采用CCK8法检测暴露于含不同浓度LPS的培养基SCAP的增殖率。通过WB和茜素红染色分别检测牙本质基质蛋白-1(DMP-1)、runt相关转录因子-2(Runx-2)和碱性磷酸酶(ALP)的表达以及矿化结节形成情况,以评估SCAP成骨/成牙本质分化情况。分别通过透射电镜和WB检测SCAP中的自噬体以及Beclin 1, 自噬相关基因-5 (Atg5)和microtubule-associated proteins light chain 3 (LC3)的表达情况。此外,检测自噬抑制剂3-methyladenine对LPS刺激下SCAP成骨/成牙本质分化的变化。结果显示,在施以5 μg/mL LPS刺激的第3天,SCAP的细胞活力减弱。LPS (5 μg/mL)抑制SCAP的成骨/成牙本质分化并减弱DMP-1, Runx-2和ALP的表达。此外,LC3, Atg5
