这一过程的第一步,研究者在卵巢切除大鼠中建立一个标准的口腔截骨模型。这种模型有2个独有的特征:第一,大鼠展现出骨质疏松的表型,相当于报道中显示的牙种植体患者人群中的骨健康水平。第二,在愈合的牙拔除部位实施截骨术,它代表着大多数植入体在患者体内的放置部位。随后,分别在一组患者和大鼠体内使用商业成品钻头实施截骨术。分子、细胞和组织学分析结果显示,人和啮齿类动物的牙槽骨对截骨部位的预备具有相近的反应。而在人和大鼠中最为显著的是,所有钻头在截骨部位周围均造成了一个骨细胞坏死和正在坏死的区域。在截骨术后收集的多个不同时间点的的大鼠组织显示,牙槽骨的最终命运均是坏死。而一周后,破骨细胞活性促使坏死骨的吸收,反过来,成骨细胞刺激了新骨基质的沉积。
