中科院苏州医工所开发出黄连素优良载体

近日,中国科学院苏州生物医学工程技术研究所检验室研究员董文飞课题组的王政等人开发出一种纳米粒子——Janus型磁介孔二氧化硅纳米粒子。利用该粒子担载黄连素,成功地提高了肿瘤细胞内黄连素的含量,并通过外加磁场的作用,进一步增强了黄连素的抗肿瘤效果。

近日,中国科学院苏州生物医学工程技术研究所检验室研究员董文飞课题组的王政等人开发出一种纳米粒子——Janus型磁介孔二氧化硅纳米粒子。利用该粒子担载黄连素,成功地提高了肿瘤细胞内黄连素的含量,并通过外加磁场的作用,进一步增强了黄连素的抗肿瘤效果。

Janus型磁介孔二氧化硅纳米粒子具有理化性质稳定、毒性低、生物相容性好、可修饰性强等优势,因此非常适合作为化疗药物的载体。黄连素是一种异喹啉生物碱,提取于中草药黄连黄柏的根茎,具有广泛的药理学作用。科学家们发现黄连素具有很好的抗肿瘤效果。然而,胃肠道吸收差、生物利用低限制了黄连素在临床上的进一步使用。

课题组通过实验,探究了Janus型磁介孔二氧化硅作为黄连素药物载体的应用价值。结果表明,研究制备的纳米粒子具有均一的形貌、高的磁性能和极好的介孔性质。在肝癌细胞和正常细胞中,空载的纳米粒子均表现了较低的毒性和极好的细胞内吞效果。利用这种纳米粒子担载黄连素,其24小时载药量高达20.7%,并呈现pH响应性释药的特征。相比于单纯的黄连素,相同剂量的磁介孔载黄连素对肝癌细胞展现了更强的杀伤作用,而对于人正常细胞,磁介孔载黄连素的毒性更低。有趣的是,在外加磁场的作用下,该纳米载药系对人肝癌细胞的抑制作用被进一步增强,而对正常细胞的作用效果不明显。研究人员通过共聚焦显微镜研究了这种现象的作用机制,发现了在人肝癌细胞中,磁介孔载黄连素组中细胞内黄连素明显多于单纯黄连素组,而外加磁场组中细胞内黄连素的量最多。而在人正常细胞中,该纳米载药系统的释放黄连素的量明显少于单纯黄连素组。这种选择性的释放解释了该纳米载药系统高效低毒的作用效果。

该实验证明了Janus型磁介孔二氧化硅纳米粒子是黄连素的优良载体,同时该研究也为利用天然中草药开发有效安全的化疗药物提供了新的思路。
