干燥综合征B细胞免疫缺陷

在SS患者中观察到的自身反应性B细胞的缺陷性反选择是许多其他自身免疫性疾病的共同特征。

在许多自身免疫性疾病患者中，中心和外周B细胞耐受检查点有缺陷，但SS患者的每个不同检查点的功能尚未评估。

使用基于PCR的方法，能够在体外克隆和表达由单个B细胞产生的重组抗体，研究人员测试了由单个CD19 + CD21low  CD10 + IgMhi CD27- 新迁移/转移克隆的重组抗体的反应性和来自5名SS患者的CD19 + CD21 + CD10 - IgM + CD27 -成熟初始B细胞。

结果发现，与健康供体相比，表达多反应性抗体的迁移性/转移性B细胞的比例显著增加，揭示SS患者的中心B细胞耐受性缺陷。在SS患者中，表达自身反应性抗体的成熟初始B细胞的比例也显著增加，从而说明这些患者中外周B细胞耐受性检查点受损。

在SS患者中观察到的自身反应性B细胞的缺陷性反选择是许多其他自身免疫性疾病的共同特征，并且可能通过允许自身反应性B细胞向T细胞呈递自身抗原而导致自身免疫的进展。
