Scientific Reports: 肝巨噬细胞耗竭可以改善间充质干细胞移植对小鼠肝损伤模型的影响

间充质干细胞(MSCs)在治疗不同类型的肝损伤时显示出不同水平的有效性,表明受损肝脏的微环境是干细胞治疗有效性的关键决定因素。因此本项研究的目的是评估肝干细胞生态成分对四氯化碳(CCl 4)诱导的肝损伤过程中移植的MSCs的调节作用。

背景:
间充质干细胞(MSCs)在治疗不同类型的肝损伤时显示出不同水平的有效性,表明受损肝脏的微环境是干细胞治疗有效性的关键决定因素。因此本项研究的目的是评估肝干细胞生态成分对四氯化碳(CCl 4)诱导的肝损伤过程中移植的MSCs的调节作用。

方法:
研究人员将超顺磁性氧化铁(SPIO)标记的人MSC静脉内注射到用CCl4处理的小鼠中并进行肝脏巨噬细胞消耗。移植后不同时间间隔收集肝组织,用于随后的组织病理学,形态学,免疫组织化学,基因表达和超微结构研究。移植的MSCs的归巢通过铁染色和免疫组织化学研究在小生境中追踪它们来证明。MSCs分化为肝细胞样细胞和内膜平滑肌细胞,这可通过它们表达人白蛋白和α-平滑肌肌动蛋白来证明,同时伴随着小鼠肝细胞生长因子水平的增加。

结论:
本项研究发现移植后肝纤维炎症反应减少,并且肝脏巨噬细胞耗尽促进了这一过程。证明了肝脏巨噬细胞耗尽可以对肝细胞治疗起到促进作用。
