Stem Cell Rep:发现关键的干细胞调节因子可以延缓小肠癌生长!

斯德哥尔摩大学(SU)的研究人员发现了果蝇小肠干细胞调节的一种新机制。此外,他们还发现一种关键蛋白质可以延缓小肠肿瘤生长。对这种机制的更深入了解可以帮助我们了解人类小肠疾病如何发生以及帮助我们开发针对小肠癌的新型疗法。这些研究结果于近日发表在《Stem Cell Reports》上。

斯德哥尔摩大学(SU)的研究人员发现了果蝇小肠干细胞调节的一种新机制。此外,他们还发现一种关键蛋白质可以延缓小肠肿瘤生长。对这种机制的更深入了解可以帮助我们了解人类小肠疾病如何发生以及帮助我们开发针对小肠癌的新型疗法。这些研究结果于近日发表在《Stem Cell Reports》上。

健康的小肠粘膜会从非免疫细胞——所谓干细胞持续产生细胞。为了与衰老损伤细胞清除的速度一样,细胞的增殖就必须被严格控制。干细胞过快增殖会导致肿瘤产生而增殖速度太慢则会导致小肠壁不正常,产生炎症。

通过探究果蝇的小肠再生,研究人员发现了小肠细胞确定它们是否继续分裂或者成熟分化为具有特定功能的特殊细胞的机制。果蝇的小肠与人类小肠有很多相似之处,因此被用于研究干细胞如何被调节以及肿瘤如何形成。

研究人员发现果蝇中一个叫做Nubbin的基因会产生两种蛋白质——Nub-PB和Nub-PD,这两种蛋白质在干细胞形成及成熟过程中的作用完全相反。正常情况下,健康小肠中的两种蛋白质处于平衡状态。当Nub-PB相对较多时,细胞会更快成熟,而当Nub-PD相对偏多时,会形成的新的干细胞,如果完全缺乏Nub-PB,这些细胞就不会成熟,就会形成肿瘤。

“我们可以把它比喻为阴和阳,中国哲学中相互作用并维持平衡的两个极端。” 来自SU分子生物科学系教授Ylva Engstrom说道。

蛋白可以延缓肿瘤生长

除此之外,研究人员还发现Nub-PB可以延缓果蝇小肠肿瘤生长。研究人员通过基因突变产生了肿瘤,但是当他们刺激增加小肠细胞Nub-PB的含量时,不会形成肿瘤。因此Nub-PB就是所谓的肿瘤抑制蛋白。

人体内也有一个相似的蛋白质,叫做Oct1,与Oct4很相关。Oct1已经被证明和人类多种癌症相关,因此某些情况下Oct1发生了突变。Oct4是一个研究透彻的干细胞因子,具有调控人细胞分裂的功能。

“这些调节因子之间平衡被打破就会造成严重的后果,导致炎症、感染甚至是癌症。”Ylva Engstrom说道。

从长远看,更深入了解这些机制将帮助我们揭开小肠疾病发生的机制并帮助我们开发预防和治疗小肠癌的新药物。
