Aging:研究人员又发现新的长寿基因

几个世纪以来,人们对能够让一些人活得格外长寿的因素一直保持着巨大的兴趣。通过敲除许多特定的基因,人们已经可以在动物模型中实现极长的寿命。在人类中,利用常规全基因组关联研究(GWAS),研究人员也已经找到了越来越多与人类长寿相关的遗传因素。

近日,来自埃克塞特大学医学院的研究人员对欧洲血统的英国生物银行参与者进行了父母长寿的全基因组关联研究(GWAS)。结合母亲和父亲的年龄,研究人员发现10个与长寿相关的基因座。其中包括8个已经被报导的基因座,和生存,阿尔茨海默病和心血管疾病等性状相关。

其中,4个基因座也与10%最长的存活率(母亲年龄≥90岁,父亲≥87岁)相关,此外,其还有两个额外的关联,包括MC2R内含子变体(编码促肾上腺皮质激素受体)。母亲的死亡年龄与另外的3个基因座有关(2个与自身免疫疾病有关),8个基因座只与父亲有关。

因此,这些结果提示,人类长寿是高度多基因的,其中可能参与细胞衰老和炎症的基因座,以及脂质代谢和心血管疾病的基因座具有突出的作用,也可能有性别特定的长寿路线。
