J Dent Res:牙龈卟啉单胞菌通过靶向GRHL2损伤口腔上皮屏障

口腔上皮是抵御多种环境和细菌刺激的第一道防线,它通过多蛋白的紧密连接(TJ)、粘附连接、桥粒和缝隙连接复合体形成上皮细胞互连屏障。Grainyhead-like 2 (GRHL2)是一种上皮特异性的转录因子,因其可调节连接蛋白的表达,可能在形成粘膜上皮屏障中起到重要作用。这篇研究的目的是为了评估GRHL2在牙龈卟啉单胞菌(Pg)诱导的上皮屏障功能损伤中的作用。

结果显示,Pg或Pg来源的脂多糖(Pg-LPS)导致人口腔角质形成细胞(HOK-16B 和OKF6 cells)内源性GRHL2和结合蛋白(如:紧密连接部、钙粘连蛋白、闭合蛋白和闭合素)的快速缺失。GRHL2直接调控结合蛋白的表达水平和小分子的上皮通透性(如:右旋糖酐和Pg菌)。为了探索GRHL2在口腔上皮屏障中的功能,研究使用Grhl2条件敲除(KO)的小鼠模型,它可诱导上皮组织特异性的Grhl2 KO。Grhl2 KO在结合上皮处损伤结合蛋白的表达并且增加结扎诱导牙周炎模型的牙槽骨缺失。荧光原位杂交结果显示,与野生对照小鼠相比较,Grhl2 KO小鼠口腔细菌的上皮通透性明显增加。同样,与同窝对照的野生型小鼠相比,Grhl2 KO小鼠的血液负载口腔细菌水平明显升高(如:类杆菌、芽孢杆菌、杆状菌、硬壁菌、b-变形菌和螺旋体)。

结论:这些结果表明,Pg菌可能部分依靠靶向口腔上皮细胞的GRHL2表达通过上皮粘膜增强细胞旁通透性,随后抑制结合蛋白表达进而损伤上皮屏障,导致牙槽组织破坏的增加和系统性菌血症。
