Sci Adv:高温或影响胎儿大脑发育

近日,中国科学院动物研究所研究员焦建伟团队发现,孕鼠经过高温处理后,胎鼠大脑皮层内神经干细胞增殖比例增加,而分化的神经元比例减少,并且会促进胎鼠大脑内一种热激活钙离子通道 TRPM2 的表达量增加。此前有报道称,TRPM2 与多种神经发育 / 神经系统疾病有关,包括躁郁症、神经性疼痛和帕金森病。相关研究发表于《科学进展》。

在怀孕过程中,许多刺激都会影响胎儿的正常发育,高温也不例外。有研究表明,妊娠期母体高温会导致胎儿出现神经管缺陷,但高温是如何影响胎儿大脑皮层发育的目前还不清楚。

近日,中国科学院动物研究所研究员焦建伟团队发现,孕鼠经过高温处理后,胎鼠大脑皮层内神经干细胞增殖比例增加,而分化的神经元比例减少,并且会促进胎鼠大脑内一种热激活钙离子通道 TRPM2 的表达量增加。此前有报道称,TRPM2 与多种神经发育 / 神经系统疾病有关,包括躁郁症、神经性疼痛和帕金森病。相关研究发表于《科学进展》。

研究人员为了研究母体高温状态下 TRPM2 和胎儿大脑皮层发育的关系,通过敲降胎鼠大脑神经干细胞内的 TRPM2 来研究其功能。结果发现在高温条件下敲低 TRPM2 会抑制胎鼠神经干细胞的增殖,神经元的发育也会受到影响,但是在正常温度的母体环境下不会引起这种表型。进一步的研究表明,在母体高体温状态下,TRPM2 会通过调控 SP5 这种转录因子影响胚胎神经发育。

“这项研究表明母体体温会调控胎儿大脑皮层的发育,明确了 TRPM2 这种热敏感蛋白参与高温状态下神经干细胞的发育调控,为预防母体高温导致的胎儿发育异常提供了新的思路。” 该论文第一作者、中国科学院动物研究所博士研究生李妍昕说。
