Biomaterials Science:制备出适用于肿瘤细胞成像和热化疗的纳米硫化铜材料

近期,中国科学院合肥物质科学研究院固体物理研究所纳米材料和纳米结构研究室研究员梁长浩课题组与哈尔滨医科大学附属第一医院教授杨秀华课题组合作,在多功能纳米硫化铜(CuS)的制备及其抗肿瘤性能研究方面取得进展,相关研究结果发表在国际期刊Biomaterials Science上。

近期,中国科学院合肥物质科学研究院固体物理研究所纳米材料和纳米结构研究室研究员梁长浩课题组与哈尔滨医科大学附属第一医院教授杨秀华课题组合作,在多功能纳米硫化铜(CuS)的制备及其抗肿瘤性能研究方面取得进展,相关研究结果发表在国际期刊Biomaterials Science上。

可视化治疗作为精准医疗的一种辅助手段,近年来在抗肿瘤治疗研究中受到广泛关注。通常的研究思路是分别合成具有成像或治疗功能的纳米材料再形成复合体系,其制备工艺相对复杂,不利于广泛推广。近年来,集成像与治疗为一体的多功能单一纳米材料,因其制备工艺简单、潜在副作用小等优点,在生物医药领域具有良好的应用前景。其中,铜的硫族化合物由于其光热转换效率高,是典型的光热治疗试剂。此外,与Gd3+和Fe2+基纳米材料类似,Cu2+具有未配对的3d轨道电子,因此,CuS纳米材料存在剩余磁矩,具有成为磁共振成像(MRI)造影剂的潜力。

基于此,梁长浩课题组与杨秀华课题组合作,采用液相激光辐照技术结合离子置换法,获得了单一组分的硫化铜花状中空纳米球(CuS HNs),并首次提出了将CuS HNs应用于磁共振成像引导的热化疗策略。作为传统的光热转换材料,合成的CuS HNs展现出了较高的光热转换效率(30%)。体外MRI成像实验结果表明,在临床颅脑检查中最常用的T1 WI和T2 FLAIR成像序列上,CuS HNs的成像亮度随样品浓度的增加而显着增强。其纵向质量弛豫速率(mr1)为5.1(mg/mL)-1·s-1,是商用T1 MRI造影剂(马根维显)的1.6倍。小鼠瘤内注射CuS HNs样品6小时后,肿瘤组织区域MRI成像亮度增加,进一步证实了CuS HNs作为T1 MRI阳性造影剂具有良好的应用前景。另外,硫化铜的三维中空花状结构具有大量的空隙和大的比表面积,可实现对抗癌药物DOX的负载,其负载量可达2.69%。活体内外研究表明,负载DOX的CuS纳米试剂不仅能实现光热治疗和药物化疗功能,而且能够结合两种功能发挥协同治疗作用,实现对肿瘤组织的高效抑制,是潜在的MRI成像引导的热化疗诊疗试剂。
