BMC Med:为什么说吃红肉致癌?这回科学家给出了分子证据

不同的饮食习惯与各种人类疾病密切相关,如癌症、心血管疾病、2型糖尿病、肥胖症和高血压。已有大量研究表明,大量食用哺乳动物源性食物(即红肉和乳制品)被认为是人类患癌症和心血管疾病的风险因素。

营养可以极大地影响健康。不同的饮食习惯与各种人类疾病密切相关,如癌症、血管疾病、2型糖尿病、肥胖症和高血压。已有大量研究表明,大量食用哺乳动物源性食物(即红肉和乳制品)被认为是人类患癌症和心血管疾病的风险因素。

近日,发表在《BMC Medicine》上的一项研究中,由以色列特拉维夫大学领导的国际研究团队发现,红肉和乳制品中存在的一种糖与血液中抗体发展之间存在直接的分子联系,而该抗体会增加患癌症的几率。该研究解释了大量食用红肉和乳制品的人群中癌症的高发病率。这与高胆固醇和心脏病风险增加之间的联系类似。

N-羟乙酰神经氨酸(Neu5Gc)是哺乳动物中常见的唾液酸类型的糖。多数食肉动物自身会合成这种分子,但人类却不能。因此,人类在婴儿期第一次接触乳制品和肉制品时,身体会产生抗Neu5Gc抗体。

饮食中的Neu5Gc被摄入后,会低水平结合到人体细胞表面,特别是在癌症中,因此显示出广泛的免疫原性Neu5Gc聚糖。事实上,到目前为止,所有接受检查的人类都有多种抗Neu5Gc抗体。因此,循环中的抗Neu5Gc抗体在人体组织上不断遇到含有Neu5Gc的抗原表位,并被认为会促进一种免疫反应和名为“xenosialitis”的慢性炎症。在小鼠中,这种情况会加剧癌症和心血管疾病的发生。

虽然已知这些抗体会增加患癌症的风险,尤其是结直肠癌的风险,但还没有发现它与摄入肉类和奶制品之间的直接联系。

在这项研究中,研究人员使用了来自NutriNet-Santé的样本,该样本是在法国进行的一项广泛的国家营养学调查。研究人员测量了法国饮食中常见的各种乳制品和肉类食品中Neu5Gc的含量,并分析了19621名18岁及以上成年人的每日Neu5Gc的摄入量。受试者在几天内在线报告了他们所有的食物摄入量。

随后,研究人员抽取了120名受试者的代表性样本,并测量了他们血液中抗Neu5Gc抗体的水平。

基于这些发现和对法国各种食品中Neu5Gc的定量分析,该研究团队建立了一个名为GCEMI的指数。该指数对那些过度食用会导致抗体增加,并可能增加癌症风险的食品进行了分类。

此外,为了评估红肉摄入量对不同国家的影响,该团队还利用联合国粮食及农业组织的数据库进行了调查。结果表明,大肠癌的发病率和死亡率均与红肉的摄入量呈正相关,且男性发病率和死亡率高于女性。这些发现与在40年前在少数几个国家进行的癌症风险的研究结果是一致的。

该研究通讯作者、特拉维夫大学乔治·怀斯生命科学学院细胞研究与免疫学系教授Vered Padler-Karavani 博士说:“我们发现从红肉和奶酪中大量摄入Neu5Gc与那些增加癌症风险的抗体之间存在显着的相关性。多年来,研究人员一直在试图寻找这样的联系。在这项研究里,由于测量血液中抗体方法的精确性和法国饮食调查问卷的详细数据,使我们第一次能找到这种分子联系。”

Padler-Karavani 博士补充说,这种方法可以让研究人员预测那些摄入大量红肉和奶酪的人会产生高水平和不同类型的抗体,因此患癌症的风险更高,尤其是结直肠癌,也包括其他癌症。
