Cell:重磅!无论抗癌,还是抗感染,强大的免疫系统需要“镁”力!

在哺乳动物细胞内,镁(Mg)是最重要的微量营养素之一,数百种酶都需要镁离子作为必需的辅助因子。镁参与人体300多种化学反应。它不仅帮助肌肉收缩,协助神经发送和接受信息,还辅助心脏稳定地跳动。此外,镁还使我们的免疫系统变得更强大。大多数人可以通过摄入绿叶蔬菜、全谷物和豆类等食物补充镁。

在过去二十年里,镁在生物体中的作用已被广泛研究,尤其是血液中的镁(血清镁)水平与免疫系统的联系。血清镁含量是免疫系统抵御病原体和癌细胞能力的一个重要因素。因此,缺乏镁与多种疾病有关,包括感染和癌症。然而,迄今为止,关于镁是如何影响免疫系统的科学家还知之甚少。

北京时间120日凌晨,发表在Cell上的一项最新研究中,来自瑞士巴塞尔大学的研究团队发现,T细胞只有在富含镁的环境下才能有效地消除异常或受感染的细胞,使免疫系统有效运作。这一发现将对癌症治疗具有重要意义。

先前的研究发现,饮食中镁摄入量低和低镁血症与各种疾病的病理生理学有关,包括感染和癌症。有研究显示,在喂食缺乏镁饮食的小鼠中,癌细胞的转移性扩散加速,并且由于诱导的T细胞激酶(ITK)活性不足,动物对流感病毒的防御能力也受到了影响。

在这些模型中,细胞外镁是否有助于形成适应性免疫反应尚未被评估。CD8+ T细胞是适应性免疫系统的重要组成部分,在识别和清除感染或恶性转化细胞方面发挥着重要作用。

在这项新研究中,研究人员发现,CD8+T细胞只有在富含镁的环境中才能有效地消除异常或被感染细胞。镁对于一种名为LFA-1T细胞表面蛋白至关重要,LFA-1T细胞激活中发挥着关键作用。

具体而言,共刺激细胞表面分子LFA-1需要镁在CD8+T细胞上采用其活性构象,从而增加钙流量、信号转导、代谢重编程、免疫突触形成,以及增强的T细胞毒性。

研究人员解释说,在非活跃状态下,LFA-1处于弯曲的结构中,因此不能有效地与感染或异常细胞结合。这正是镁发挥作用的地方。如果在T细胞附近有高水平的镁,它就会与LFA-1结合,并确保其保持活性。

镁对T细胞的功能至关重要,这一事实可能是现代癌症免疫疗法的一个非常重要的发现。这些疗法的目的是调动免疫系统(尤其是细胞毒性T细胞)来对抗癌细胞。在实验模型中,研究人员证实,T细胞对癌细胞的免疫反应因肿瘤中局部镁水平的增加而得到加强。

通过分析之前的癌症患者临床数据,研究人员证明,免疫疗法对血清镁水平不足的患者效果较差。

总之,这些发现在概念上将共刺激和营养感知联系了起来,并指出-LFA-1”轴是一个治疗性的生物系统。

研究通讯作者、巴塞尔大学生物医学系Christoph Hess教授说:根据现有数据,我们还不能回答经常摄入镁是否会影响患癌症风险的问题。下一步,我们计划进行前瞻性研究,以测试镁作为免疫系统催化剂的临床效果。
