Med Sci Monit:钛表面应用锶负载的相变溶菌酶涂层增强成骨和骨免疫调节作用

通过新型的一步相转移溶菌酶(PTL)处理来制备掺锶(Sr)的钛(Ti)表面,并研究制备的样品对成骨和骨免疫调节的影响。

通过新型的一步相转移溶菌酶(PTL)处理来制备掺锶(Sr)的钛(Ti)表面,并研究制备的样品对成骨和骨免疫调节的影响。

 

分别制备了5组钛标本,包括Ti、PTL、PTL@10Sr(PTL包被10 mg/mL Sr)、PTL@20Sr PTL包被20 mg/mL Sr)和PTL@50Sr(PTL包被50 mg/mL Sr)组。通过免疫荧光、MTS检测和Transwell系统,对骨髓间充质干细胞(BMSCs)在不同表面上的初始附着、扩散、增殖和迁移等行为进行了研究。然后检测了BMSCs的成骨性分化。通过实时PCR检测巨噬细胞诱导的表面的极化,并评估BMSCs对巨噬细胞培养基的反应,从细胞迁移和成骨分化的角度进行评估。最后,我们进行了一项体内研究,利用大鼠股骨移植模型,评估材料的成骨诱导和成骨免疫调节潜力。

 

体外实验表明,PTL涂层可以改善细胞的扩散和粘附,PTL@Sr层稳定释放的Sr可以促进细胞迁移和成骨。此外,PTL@Sr表面可以调节巨噬细胞的免疫反应,增强BMSCs的募集和成骨分化。体内评价显示,PTL@20Sr组的炎症浸润减少,骨形成改善。

 

综上所述,该研究结果表明,Sr负载的PTL层对诱导BMSCs成骨分化有更大的潜力,同时Sr负载的PTL层可以调节免疫反应,从而在体外和体内促进成骨。
