研究表明,具核梭杆菌(F. nucleatum)在结直肠癌患者的肿瘤组织和粪便样本中富集,诱导肿瘤细胞大量产生miR-1246/92b-3p/27a-3p和CXCL16/RhoA/IL-8富集的外泌体

Science 杂志发表了一篇重磅论文。以色列魏茨曼科学研究所的科研团队发现,在人类的肿瘤细胞里,存在许多细菌。

早在100多年前,科学家们就在人类的肿瘤里检测出了细菌,一些人认为,这表明细菌在肿瘤的微环境里可能具有局部的作用;另一些人则指出,这些细菌的含量都非常低,很难确认它们真的来自肿瘤样本,还是来自外部的污染。

这篇 Science 论文分析了乳腺癌、肺癌、黑色素瘤、胰腺癌等常见的7种实体肿瘤,样本数超过了1500份。分析结果确认,大部分的肿瘤样本里都含有细菌。而且,这些细菌大多数都位于肿瘤细胞内!但是这些细菌在肿瘤中具体作用却并不是很清楚。

2020年11月11日,中山大学药学院张革教授团队在 国际著名学术期刊 Gut(IF=19.82)上发表了题为:Exosomes derived from Fusobacterium nucleatum-infected colorectal cancer cells facilitate tumor metastasis by selectively carrying miR-1246/92b-3p/27a-3p and CXCL16 的研究论文。

该研究表明,具核梭杆菌(F. nucleatum)在结直肠癌患者的肿瘤组织和粪便样本中富集,诱导肿瘤细胞大量产生miR-1246/92b-3p/27a-3p和CXCL16/RhoA/IL-8富集的外泌体,并进一步促进癌症转移。

该研究开拓了瘤内感染性细胞特异性外泌体这一新领域的研究,为深入解析致癌性细菌-宿主相互作用分子机制提供了新的见解,也为临床结直肠癌治疗提供了有价值的思路。

张革教授团队前期研究发现具核梭杆菌是兼性胞内菌,能在巨噬细胞中存活并增殖,是首个被报道的严格厌氧的胞内菌,开拓了严格厌氧菌宿主细胞内存活研究的新领域。

胞内菌感染可以介导宿主细胞分泌性外泌体(exosome)的改变,外泌体通过其携带的蛋白质、核酸、脂质和代谢物等发挥细胞间通讯功能,参与多种生理和病理过程。近年来,肿瘤细胞分泌的外泌体在癌症中的作用被明确和解析,瘤内细菌感染性外泌体的分泌及其对肿瘤的作用还是一新领域。

该项研究通过多组学技术手段发现:具核梭杆菌感染结直肠癌细胞后,诱导宿主细胞后大量产生miR-1246/92b-3p/27a-3p和CXCL16/RhoA/IL-8富集的外泌体,这些肿瘤兼具感染特异性的外泌体被传递到微环境中未感染的结直肠癌细胞中,通过miRNA和蛋白多个靶分子和多个作用模式共同从而促进了结直肠癌的转移。

这项研究研究开拓了瘤内感染性细胞特异性外泌体这一新领域,为深入解析致癌性细菌-宿主相互作用分子机制提供了新的见解,提示了具核梭杆菌感染结直肠癌细胞来源的外泌体可能是一种重要的致癌因素促进结直肠癌的发展和转移,并指出肿瘤治疗中瘤内具核梭杆菌清除的重要性,为临床结直肠癌治疗提供了有价值的思路。

中山大学药学院博士生郭松鹤为第一作者,硕士生陈俊、陈芳芳为第二和第三作者,张革教授为通讯作者。本研究得到了国家自然科学基金的支持及中山大学肿瘤防治中心刘万里教授课题组的帮助。
