器官的功能和再生能力会随着衰老或成年干细胞系统衰竭而衰竭。干细胞分裂在多细胞生物衰老/应激细胞的命运和动力学中的作用尚不清楚。干细胞有两种分裂形式,一种是对称细胞分裂(SCD),通过将有丝分裂纺锤体定向为平行于基础基底膜来生成具有相同命运的两个细胞;一种是非对称细胞分裂(ACD),通过将有丝分裂纺锤体定向为垂直于基础基底膜而生成自我更新的干细胞和分化细胞。

毛囊(HF),是哺乳动物长出毛发的微器官,在衰老过程中会变细,导致头发稀疏和脱落。既往研究表明,条件性敲除基底细胞的细胞极性蛋白aPKCλ可增加ACD、废除SCD,导致毛囊干细胞(HFSC)耗竭而导致小鼠脱毛。

衰老的HFSC的分裂

但在衰老过程或应激状态下,HFSC则通过非典型的“应激响应”型非对称细胞分裂,这种分裂伴随着半桥粒蛋白COL17A1和细胞极性蛋白aPKCλ的失稳性。具体而言,衰老的HFSC发生有丝分裂的频率明显降低,而且明显丧失了细胞极性相关蛋白的极性分布或表达

通过在部分HF中的垂直细胞分裂,衰老的HFSC优先在顶侧优先生成KRT10 +表皮角质形成细胞,而不是KRT75 + HF角质形成细胞。总之,衰老的HFSC会自然激发非典型的ACD,直接产生终末分化的表皮细胞,而不是具有再生能力的毛囊生态位。随着这种非典型的垂直分裂的不断重复,HFSC从逐渐基底膜脱离,导致基底膜的HFSC耗竭、消失,乃至器官衰老,最终导致毛囊收缩和头发稀疏。

通过aPKCλ诱导COL17A1可恢复HFSC功能

干细胞分裂方向可以通过外部因素(如β1-整合素)或内部细胞极性蛋白aPKCλ(调节单层上皮细胞SCD过程中的纺锤体定位)调控。通过aPKCλ的稳定作用,实验性诱导COL17A1的稳定性可拯救器官的体内稳态
