此外,脂质转运已被证实可调节胰岛素的分泌,而广泛的表型关联研究(PheWAS)和孟德尔随机化(MR)方法均发现CAD风险升高相关的单核苷酸多态性(SNP)与LDL的水平升高相关,而空腹胰岛素水平和T2D风险降低相关。


既往研究显示,LDL、CAD和T2D之间的关系在药理学上也已得到很好的建立。通过降脂药物降低血浆LDL的水平不仅会导致CAD相关临床事件和冠状动脉粥样硬化斑块中脂质含量的降低,而且还可能增加T2D的风险。

大量的大型队列研究表明,他汀类药物疗法在产生重大血管益处的同时,也会增加T2D的患病风险。然而,他汀类药物的多种生化作用尚未在分子水平上得到充分表征。


因此,研究人员假设,将脂质和葡萄糖调节置于更广泛的分子环境中的全面转录组模型对于揭示脂质水平、CAD和T2D之间看似矛盾的联系的重要性。

差异表达分析显示与葡萄糖和脂质代谢相关的基因和模块

在该研究中,研究人员通过STARNET(斯德哥尔摩-塔图动脉粥样硬化逆向网络工程团队)研究这一迄今为止最大的人类数据集,采用多尺度的方式构建综合网络模型,旨在研究多元组学和多组织框架下的CAD和相关的心脏代谢紊乱。


研究人员从CAD患者的冠状动脉旁路移植术中获得七个心脏相关代谢组织并构建分子网络模型:主动脉根(AOR)、乳腺动脉(MAM)、肝脏(LIV)、皮下脂肪(SF)、内脏脂肪(VAF)、骨骼肌(SKLM)和全血(Blood),以评估脂质和葡萄糖代谢的分子成分。

GLD网络中基因表达与糖脂代谢状态的关系

通过整合基因表达、基因型、代谢组学和临床数据,研究人员确定了高度保守的葡萄糖和脂质(GLD)调控网络,其是提供相关因果调节联系的最一致的网络模型,该网络也定义了脂质和葡萄糖代谢之间的逆向关系,即脂质和葡萄糖代谢成负相关。


GLD网络的主调节因子也会逆向影响脂质和葡萄糖水平。在小鼠模型中,对GLD网络主调节因子之一的LSS(羊毛甾醇合酶)的抑制作用证实了之前所预测的葡萄糖和脂质水平的负相关关系,并提供了机制上的见解。


总而言之,该研究确定了一个逆向调节脂质和葡萄糖代谢的小型基因网络,并强调了这种多尺度、综合网络方法的能力,其能够揭示新型基因相互作用,而该作用是血浆脂质和血糖代谢双重调控的基础。
