Nat Aging:杂志首文:Sestrin是膳食影响干细胞功能和寿命的关键调节因子

最近,我国研究人员在Nature Aging杂志发表了首篇长文,阐述了Sestrin无效突变体果蝇对DR的寿命反应钝化。

已有的研究显示,饮食限制(DR)可以延长不同物种的寿命,包括果蝇和啮齿动物。DR不仅可以增加寿命,而且在啮齿类动物,灵长类动物和人类的老化过程中可以诱导广泛的健康改善。
限制特定的饮食成分,特别是蛋白质,比整体减少热量更重要。此外,限制特定的必需氨基酸,包括蛋氨酸和支链氨基酸(BCAAs)亮氨酸,异亮氨酸和缬氨酸,可以延长寿命。然而,这些现象背后潜在的分子和细胞机制仍然难以捉摸。

进化保守的Sestrin蛋白,是雷帕霉素复合物1(TORC1)靶标活性的抑制剂,最近被发现为体外氨基酸的传感器。最近,我国研究人员在Nature Aging杂志发表了首篇长文,阐述了Sestrin无效突变体果蝇对DR的寿命反应钝化。
在一个突变的Sestrin蝇系中,Sestrin与氨基酸结合能力受阻,TORC1被激活,表现出发育延迟,生育能力降低,寿命延长,对缩短寿命的高蛋白饮食不敏感。

Sestrin通过调节TOR途径和自噬,介导DR降低肠道干细胞活性和肠道细胞周转率,以及膳食氨基酸诱导的干细胞增殖。肠道干细胞中Sestrin的表达足以维持肠道稳态,延长寿命。
因此,Sestrin是膳食氨基酸、干细胞功能和长寿之间的分子联系。
