大量的证据表明了循环RNAs(circRNAs)在调控癌症发病机理方面起着关键的作用。然而,circRNAs在膀胱泌尿肿瘤(UCB)中的机制仍旧有很多不清楚。最近,有研究人员发现circFAM114A2在UCB组织样本和细胞系中均显著下调,并且表达水平与病理TNM阶段和等级显著相关。功能上讲,circFAM114A2的过表达能够显著的抑制体外UCB细胞的迁移、浸润和增殖,并抑制体内肿瘤的生长。同样

最近,有研究人员发现circFAM114A2在UCB组织样本和细胞系中均显著下调,并且表达水平与病理TNM阶段和等级显著相关。功能上讲,circFAM114A2的过表达能够显著的抑制体外UCB细胞的迁移、浸润和增殖,并抑制体内肿瘤的生长。同样的,研究人员确认了miR-762在5637和T24细胞系中被circFAM114A2大幅度的下调。荧光原位杂交(FISH)进一步的明确了circFAM114A2与miR-762之间存在细胞质相互作用。通过使用荧光素酶报告试验,研究人员发现miR-762能够直接的靶向TP63。之后,研究人员发现circFAM114A2能够通过作用miR-762来增加ΔNP63(UCB中TP63的一种主要同种型)的表达。

最后,研究人员指出,他们的结果阐释了circFAM114A2在调控ΔNP63表达中是miR-762的一种竞争性内源RNA(ceRNA),从而通过circFAM114A2/miR-762/ΔNP63信号轴来抑制UCB的进展。
