重组人GM-CSF治疗COVID-19住院患者,SARPAC临床试验达到了肺功能改善的主要终点

sargramostim并不会加重或诱发细胞因子风暴,反而可以避免它。

制药公司Partner近日公布了SARPAC研究的最新结果,该研究旨在评估酵母衍生的重组人GM-CSF Leukine®(sargramostim)治疗COVID-19住院患者的有效性和安全性。这项前瞻性、随机、开放标签研究(NCT04326920)由比利时根特大学领导,在比利时的五家医院中进行。该研究招募了81例经PCR确诊的COVID-19患者,他们患有需要补充氧气的急性低氧性呼吸衰竭。

结果显示,与单独使用标准护理(SOC)相比,吸入型sargramostim与SOC联合治疗五天后患者氧合得到显著改善。sargramostim与SOC联合治疗组有54%的患者氧合得到改善(通过肺泡动脉氧梯度测量,定义为至少比基线高33%以上),而SOC单独治疗组有26%的患者氧合得到改善(p = 0.0147)。

Sargramostim是一种细胞因子,该研究的另一个目的是观察Sargramostim在易发生细胞因子风暴的COVID-19患者中是否安全。Sargramostim组患者会经历更多的鼻出血(20%比5%),这些出血大多是轻度的(1级和2级)和短暂的。接受Sargramostim的患者的关键炎症标志物水平稳定或下降。这些发现表明,sargramostim并不会加重或诱发细胞因子风暴,反而可以避免它。
