刺芒柄花素抑制宫颈癌的机制研究,为进一步研究和开发宫颈癌抗癌药物提供依据

宫颈癌是女性与癌症相关死亡中最常见的癌症。目前,宫颈癌的治疗主要包括手术和同步放化疗。然而,宫颈癌的治疗效果仍然很差,5年生存率约为40%。因此,迫切需要开发新的治疗策略以更有效的治疗宫颈癌。

目前,PD-1/PD-L1免疫检查点由于对包括宫颈癌在内的多种癌症的免疫应答具有调节作用,已成为潜在的治疗靶点。PD-1表达于淋巴细胞,特别是T细胞,而PD-L1表达于T细胞和抗原提呈细胞。PD-L1和PD-1结合会破坏淋巴细胞的效应功能,增强抗病毒T细胞的长时间阻滞。这些变化导致免疫活性受损和T细胞衰竭。然而,除了已知的对免疫反应的影响外,PD-L1的表达在肿瘤细胞本身也发挥着固有的作用,它作为“分子盾牌”保护癌细胞免受细胞溶解。目前为止,许多研究表明宫颈癌与PD-L1过表达有关。因此,PD-L1抑制剂被认为是宫颈癌的有效治疗方法

原癌基因MYC诱导细胞增殖、生长和分化,MYC诱导的肿瘤依赖于MYC的持续表达。研究证据表明MYC是PD-L1表达的重要调控因子。在STAT蛋白中,STAT3在调节细胞增殖、存活和分化中起重要作用。证据表明,STAT3的激活增加了PD-L1表达。黄芪药食同源,具有抗炎、抗氧化、保肝作用。刺芒柄花素是从黄芪中提取的一种异黄酮,可增强细胞周期阻滞,抗血管生成,并可在一系列肿瘤,如宫颈癌、肺癌、结直肠癌和乳腺癌中转移。之前的研究表明,刺芒柄花素对多种癌细胞具有抑制能力。然而刺芒柄花素的抗肿瘤作用机制尚不清楚

近日在Journal of Nutritional Biochemistry发表的一项研究,通过体内和体外实验,利用Discovery Studio 2017/CDOCKER进行分子对接实验,探索刺芒柄花素与PD-L1、STAT3和MYC蛋白的结合模式;免疫共沉淀、Western blot/RT-PCR检测RAF、STA T3、JAK2、Src、MEK、p70S6K、ERK、MMP-9、VEGF、ICAM-1、JAK1、p-JAK2、PD-L1、MYC、p-STAT3等蛋白/RNA的表达水平组织病理学、免疫组化进行体内验证,探索刺芒柄花素对宫颈癌的体外和体内抗肿瘤作用。

结果表明,刺芒柄花素通过降低MYC和STA T3蛋白的表达来抑制PD-L1蛋白的合成。刺芒柄花素通过RAS/ERK信号通路显著降低MYC蛋白的表达,通过JAK1/STAT3信号通路抑制STAT3的激活。免疫共沉淀实验表明刺芒柄花素通过干扰MYC与STAT3的相互作用抑制PD-L1蛋白的表达。刺芒柄花素通过TFEB和TFE3介导的溶酶体发生促进PD-L1蛋白降解。T细胞杀伤实验表明刺芒柄花素能增强细胞毒性T淋巴细胞(CTLs)的活性,恢复对肿瘤细胞的杀伤能力。刺芒柄花素通过抑制PD-L1抑制细胞增殖、管形成、细胞迁移,促进肿瘤细胞凋亡。最后,在小鼠异种移植瘤模型中证实了芒柄花素对肿瘤生长的抑制作用。

该研究揭示了刺芒柄花素的抗肿瘤作用,为进一步研究和开发宫颈癌抗癌药物提供了依据
