摘要:

虽然骨髓(BM)壁龛细胞对于造血干细胞(HSC)的维持至关重要,但应激时,他们之间的相互作用尚不明确。Ramasz等人利用急性血小板减少症的小鼠模型来研究HSCs和壁龛细胞在血小板池恢复过程中的相互作用。

一过程需要巨核细胞膜定位的干细胞因子(m-SCF)的参与,反过来m-SCF又受血管内皮生长因子A (VEGF-A)和血小板衍生生长因子B (PDGF-B)的调控。随后,HSCs和多能祖细胞2 (MPP2),而非MPP3/4,被一种依赖于RTK的双受体酪氨酸激酶信号事件(即m-SCF/c-Kit和VEGF-A/VEGFR-2)激活,进而促进了它们的选择和早期增殖。

综上所述,本研究结果描述了一个单一血细胞急性丢失所引发的动态信号网络,并揭示了三种RTKs及其配体,在血小板减少症中,协调造血干细胞和祖细胞选择性激活方面的重要作用。
