Nat Genet:研究发现浸润性小叶癌新型驱动基因

浸润性小叶癌 (Invasive lobular carcinoma, ILC)是乳腺癌中常见的第二大亚型,占乳腺癌总体病例的8-14%。尽管在人体中,大多数浸润性小叶癌以上皮细胞钙黏蛋白(CDH1)的功能缺失为特征,小鼠模型中Cdh1的失活并不会使其更容易患乳腺肿瘤,这就说明在小鼠中,浸润性小叶癌的形成还需要积累其他基因上的突变。

为了明确这些基因,研究人员采用”睡美人”转座子系统(Sleeping Beauty transposon system)在乳腺特异性Cdh1失活的小鼠中,展开插入诱变筛查。这些小鼠发展出了多种独立的乳腺肿瘤,根据形态学和基因表达的分析,他们之中的大多数都和人类中浸润性小叶癌相似。

另外,在Myh9, Ppp1r12a, Ppp1r12b 和Trp53bp2中发现了周期性且相互排斥的转座子插入,其产物参与了肌动蛋白细胞骨架的调节。值得注意的是,Myh9, Ppp1r12b 和Trp53bp2也常出现在人类浸润性小叶癌中,突出了这些基因在浸润性小叶癌发展进程中,新型致癌通路中的驱动作用。
