CELL:癌基因ALK竟也帮助瘦子吃不胖

最近,研究人员在一个表型独特的爱沙尼亚队列中,对代谢健康的瘦人(全人群BMI谱的最低6个百分位数)进行了GWAS研究。

尽管世界上大部分地区的肥胖环境都是同样的,但在体重增加的易感性方面存在着相当大的个体间差异。

尽管已有的许多研究的重点是确定肥胖的遗传易感性,但最近,研究人员在一个表型独特的爱沙尼亚队列中,对代谢健康的瘦人(全人群BMI谱的最低6个百分位数)进行了GWAS研究。

研究人员发现了一个候选的瘦人基因---非细胞增生性淋巴瘤激酶(ALK)。在果蝇中,RNAi介导的ALK敲除导致甘油三酯水平的下降。在小鼠中,ALK的基因删除导致动物体型瘦小,并对饮食和瘦素突变诱导的肥胖有明显的抵抗力。

从机制上讲,研究人员发现下丘脑神经元中ALK的表达通过交感神经控制脂肪组织溶脂的能量支出。

因此,该遗传学和机理实验发现ALK是一种瘦身基因,参与抵抗体重增加。
