炎症性疾病多发性硬化(MS)及其动物模型实验性自身免疫性脑脊髓炎(EAE)的特点是外周免疫细胞浸润到中枢神经系统(CNS),最终导致大脑和脊髓的脱髓鞘和轴突损伤。MS/EAE中的免疫细胞浸润是局部血脑屏障(BBB)破坏的结果,它使外周细胞和分子无法控制地流入。BBB是一种受严格调控的血管屏障,由特殊的内皮细胞(ECs)组成,通过细胞间紧密连接和粘附连接连接,从而控制细胞和分子在BBB上的流动。在MS和EAE中,这些连接蛋白的紊乱和细胞粘附分子(如细胞间粘附分子-1(Icam-1)和血管粘附分子-1(Vcam-1)的上调与外周细胞粘附和迁移到中枢神经系统有关。促炎细胞因子白细胞介素1 (IL-1)与MS及EAE的发病机制密切相关。

在本文中,我们使用了缺失其信号受体IL-1R1的小鼠来研究IL-1信号在BBB- ECs、星形胶质细胞和小胶质细胞中对EAE发育的作用。我们发现小胶质细胞和星形胶质细胞中的IL-1信号在EAE的发生中是多余的,而BBB-ECs中IL-1R1的缺失显著改善了疾病的严重性。BBB-ECs中IL-1信号上调粘附分子Vcam-1、Icam-1和趋化因子受体Darc的表达,所有这些都曾被证明能促进CNS特异性炎症反应。相反,IL-1R1信号抑制BBB-ECs中应激反应性血红素分解代谢酶血红素氧合酶-1 (HO-1)的表达,通过与IL-1应答基因Vcam1、Icam1和Ackr1(Darc)的调控表达失调的相关机制促进疾病进展。从机制上讲,我们的数据强调了BBB-EC IL-1信号和HO-1的功能性串扰,控制下游促炎基因的转录,从而促进自身免疫性神经炎症的发病机制。

方法:为了分离CNS-EC,按照上述方法处死小鼠并经心灌注。解剖的CNS组织用含有40μg/ml DNase I的2mg/ml木瓜蛋白酶(Sigma-Aldrich)溶液在37℃下消化30分钟。在孵育过程中,使用温和的MACS解离仪(Miltenyi)对组织进行机械匀浆。所得细胞悬浮液通过70μm细胞过滤器过滤,并在15°C下以20%Percoll梯度离心30分钟,300×g。含有CNS细胞混合物的颗粒用于流式细胞术染色。

总而言之,我们的研究支持NF-κB和Nrf2/HO-1通路之间的功能性串扰。然而,确定有效和特异的Nrf2激活剂或其抑制剂靶向治疗MS和其他炎症性疾病需要进一步的详细研究。我们相信,对这种串扰的精确理解将有助于靶向和操纵Nrf2和NF-κB信号之间的平衡,这最终代表了改善MS治疗的一个有希望的途径。
