JCLA:评价两种实时PCR检测分枝杆菌的性能

研究人员采用Anyplex试剂盒对143例呼吸道标本进行常规PCR检测。在6个月和1个月的时间里,分别连续采集87例阳性样本(65MTB22NTM)56例阴性样本。在常规支原体检测和常规PCR检测的基础上,对PCR检测方法(AdvanSurePowerChek)诊断效果进行回顾性分析。

从培养结果来看,AdvanSurePowerChekMTB检测的敏感性/特异性分别为90.7%/87.6%92.6%/85.4%。对于PCR阳性标本,涂片阴性标本的定量循环(Cq)值高于涂片阳性标本(P < 0.001)。结果表明,两种PCR方法对NTM的检测灵敏度相同,均为90.0%,特异性分别为99.2%98.4%。三种PCR检测方法的总符合率,MTB96.5%NTM97.9%

在我们的研究中,PCR检测的敏感性可能被高估了,因为该研究纳入了相对较少的PCR阴性样本,这些样本可能错过了PCR阴性但培养阳性的样本。然而,用于检测MTBNTM的两种实时PCR检测方法在相对性能评估中表现相同,其Cq值可认为适用于预测涂片阳性标本。
