Aging Cell:α-酮戊二酸可延缓年龄增长带来的生育力下降

在最近的一项研究中,来自中国农业大学的研究人员考察了α-酮戊二酸(α-KG)与小鼠、猪和人类等哺乳动物生殖衰老的潜在关联。人类卵泡液中的α-KG水平随衰老有明显的降低趋势。

由于现代人生育年龄的推迟,延长有效生育年龄成为女性的普遍要求。卵巢在哺乳动物的生殖中起着核心作用。卵巢的保留和卵母细胞的能力随着年龄的增长而下降,这也被称为卵巢衰老(同义词:生殖衰老),通常与生育力下降有关。

卵巢衰老比一般衰老发生得早得多。卵巢衰老最明显的特征是卵母细胞的质量和数量显著下降。例如,一个人类女性出生时大约有40万个带有初级卵母细胞的原始卵泡。这些初级卵母细胞一直停滞在减数第一次分裂的前阶段,直到性成熟。经期开始后,平均每月会丢失1000个初级卵母细胞,到了中年(37岁左右),一个女性的卵子只剩下25000个左右。因此,保存卵泡库,保持卵母细胞质量,将延缓卵巢衰老,延长女性生育期。

在最近的一项研究中,来自中国农业大学的研究人员考察了α-酮戊二酸(α-KG)与小鼠、猪和人类等哺乳动物生殖衰老的潜在关联。人类卵泡液中的α-KG水平随衰老有明显的降低趋势

为了探讨其机制,研究人员选择小鼠作为动物模型。α-KG可以抑制ATP合成酶,能量代谢的改变引发营养传感器下调mTOR通路。这些事件不仅有利于一般的衰老过程,还能维持卵巢功能,延缓生殖功能衰退。

考虑到α-KG作为能量代谢中天然存在的分子的安全性,其在包括人类在内的大型哺乳动物生殖中的效用值得进一步研究
