新冠肺炎密切接触者,在暴露后4天内使用羟氯喹不能降低其感染风险

近日研究人员考察了羟氯喹在SARS-CoV-2暴露后的症状性感染预防效果。
 
研究在美国和加拿大开展,为一项随机、双盲、安慰剂对照试验,测试羟氯喹作为暴露后预防措施的效果。研究人员招募了成年志愿者,其暴露于确诊患者存在的家庭或职业环境,与感染者的距离不足6英尺,暴露时间超过10分钟,且在接触时未佩戴口罩或护目镜 (高暴露风险),或者且佩戴口罩但不佩戴护目镜(中等风险暴露)。在暴露后4天内,随机服用安慰剂或羟氯喹(800毫克一次,随后在6至8小时内服用600毫克,然后在4天内每天服用600毫克)。研究的主要结果是14天内实验室确诊的Covid-19感染。
 
821名无症状参与者。总体而言,87.6%的参与者报告说,与Covid-19患者有高风险接触。羟氯喹治疗组(414例中的49例[11.8%])和安慰剂组(407例中的58例[14.3%])新冠肺炎发病率没有显著差异;绝对差异为-2.4%。羟氯喹的副作用更常见(40.1%vs16.8%),但没有严重的不良反应报告。
 
研究认为,新冠肺炎密切接触者,在暴露后4天内使用羟氯喹不能降低其感染风险。
