Nat Aging:靶向表观遗传酶KDM4,帮助治疗衰老和肿瘤

最近,研究人员发现,组蛋白H3特异性去甲基化酶KDM4在人类基质细胞发生衰老时被表达。在临床肿瘤学中,KDM4的上调以及H3K9/H3K36甲基化的减弱与前列腺癌患者化疗后的生存率较差相关。

已有的研究表明,衰老是一种自我控制机制,通过实施稳定的、持久的、通常是不可逆的生长停滞来限制细胞的过度增殖,并防止肿瘤的发展。衰老的细胞在核和染色质结构、表达模式和代谢活动方面显示出深刻的变化。衰老细胞还积极分泌大量的蛋白质,其中许多本身就是促炎因子,这种特性被统称为SASP。

一些证据表明,SASP对组织修复、伤口愈合、胚胎发育和清除衰老细胞的免疫监视至关重要。然而,作为一个进化的多面性问题,这种表型在许多病理环境中大多是有害的。例如,在局部肿瘤微环境(TME)中,残留的衰老细胞的分泌组可以明显地促进邻近癌细胞的恶变,特别是化疗抵抗,并导致许多年龄相关的疾病中的慢性炎症。

尽管有限的疗效促进结果取决于特定的环境,但SASP在晚期癌症中的负面影响远远超过其有益的贡献,并最终加速了疾病的发展。具体来说,SASP带来的有害炎症表明,消除衰老细胞或抑制SASP对包括癌症在内的多种类型的病症都是有利的。

研究人员通过对转座酶可及染色质的高通量测序,绘制了染色质可及性的全面图谱,并通过RNA测序进行表达谱分析,揭示了染色质开放性的整体变化和转录组景观的时空重编程,这是衰老相关的分泌表型(SASP)的基础。

细胞衰老时,组蛋白H3赖氨酸位点的表观遗传变化

研究结果显示,选择性靶向KDM4可以抑制衰老基质细胞的SASP,促进治疗破坏的肿瘤微环境中的癌细胞凋亡,并延长了实验动物的生存期。

该研究支持衰老过程中H3K9/H3K36甲基化的动态变化,确定了一种异常容许的染色质状态,并揭开了KDM4作为SASP关键调节因子的面纱。

总之,该研究以KDM4为靶点,提出了一个新的治疗途径,以操纵细胞衰老并限制其对包括癌症在内的年龄相关病症的贡献
