Cell:人类乳腺癌的肿瘤和免疫生态系统单细胞图谱

乳腺癌是一种异质性疾病。肿瘤细胞和相关的健康细胞形成决定疾病进程和对治疗反应的生态系统。为了表征乳腺癌生态系统的特征及其与临床数据的关系,我们使用质谱分析了144例人类乳腺肿瘤和50例非肿瘤组织样本。

乳腺癌是一种异质性疾病。肿瘤细胞和相关的健康细胞形成决定疾病进程和对治疗反应的生态系统。为了表征乳腺癌生态系统的特征及其与临床数据的关系,我们使用质谱分析了144例人类乳腺肿瘤和50例非肿瘤组织样本。使用肿瘤和以免疫细胞为中心的抗体组评估了2,600万个细胞中73种蛋白质的表达。

 

结果显示,肿瘤在肿瘤细胞组成中表现出个性,包括表型异常和表型优势。肿瘤与免疫细胞之间的关系分析揭示了与免疫抑制和预后不良有关的生态系统特征。在高等级的ER +和ER-肿瘤中发现高频率的PD-L1 +肿瘤相关巨噬细胞和疲惫的T细胞。

 

总之,这种大规模的单细胞图谱加深了我们对乳腺肿瘤生态系统的了解,并表明基于生态系统的患者分类将有助于识别针对肿瘤及其免疫环境的精密医学方法的个体。
