肝细胞癌(Hepatocellular carcinoma , HCC)是全球范围内死亡率极高的恶性肿瘤之一,治疗效果及预后极差。RNA结合蛋白(RNA banding proteins, RBPs)介导的转录后调控在许多生理和病理进程中发挥重要作用。然而,大多数RBPs在HCC进展中的生物学功能和机制尚不清楚。

肝细胞癌(Hepatocellular carcinoma , HCC)是全球范围内死亡率极高的恶性肿瘤之一,治疗效果及预后极差。RNA结合蛋白(RNA banding proteins, RBPs)介导的转录后调控在许多生理和病理进程中发挥重要作用。然而,大多数RBPs在HCC进展中的生物学功能和机制尚不清楚。

近日,北京大学衰老研究中心童坦君课题组与北京协和医院合作,就RNA结合蛋白RPS3在肝细胞癌(Hepatocellular carcinoma , HCC)中的表达谱及对HCC肿瘤进展的影响和分子机制方面的研究成果进行了系统阐述。同时,研究还发现了一种新的RPS3抑制剂——4-羟基-2-亚甲基丁酸-(5-甲酰基呋喃-2-甲)酯(缩写为FMHM),该小分子化合物从保健药品中提取,能够通过抑制RPS3/SIRT1途径抑制HCC进展。相关成果以“RNA-binding protein RPS3 contributes to hepatocarcinogenesis by post-transcriptionally up-regulating SIRT1”为题在Nucleic Acids Research杂志发表。


FMHM通过抑制RPS3/SIRT1途径抑制HCC进展

该工作通过生物信息学分析TCGA/LCI/GEO数据库中共887个HCC临床样本的测序信息,成功鉴定了一组肝癌进展相关的RNA结合蛋白(HPARBPs),并利用免疫组化芯片分析了RPS3在人HCC样本中的表达谱,通过表达和敲低HCC细胞模型及裸鼠模型中RPS3的表达,研究RPS3对HCC肿瘤进展的影响。经RNA-seq寻找RPS3发挥作用的下游靶基因,课题组通过RNA-pull-down及RNA-IP实验进行多方面的验证,在细胞水平评估了FMHM对RPS3&SIRT1的影响,并在动物水平评估了FMHM的治疗价值。该项研究成果表明,RNA结合蛋白RPS3是肝癌发生中的重要促肿瘤发生因子,能够通过转录后调控SIRT1的表达影响HCC进展。此外,RPS3可能成为筛选肝癌药物的药物靶点,FMHM靶向调控RPS3/SIRT1通路可能为肝癌的的临床治疗提供新的方向。
