该MR研究提供了支持证据，表明吸烟者终生吸烟与乳腺癌风险之间存在潜在的因果关系，但每天吸烟量与其无关。

尽管根据最近的流行病学研究报告，吸烟和患乳腺癌风险之间有一定的联系，但吸烟是否与乳腺癌风险之间存在因果关系仍未明确。

近日，肿瘤学领域权威杂志British Journal of Cancer上发表了一篇研究文章，研究人员应用孟德尔随机化(MR)来评估吸烟与乳腺癌风险之间潜在的因果效应。

在终生吸烟指数(LSI)或每天吸烟(CPD)的全基因组关联研究中报告了164个单核苷酸多态性(SNPs)的个体水平数据和汇总统计数据，它们被用于获得MR效应估计值。来自于108420例浸润性乳腺癌患者和87681例对照者的数据被用于LSI分析，并且在26147例癌症患者和26072例对照者中进行了CPD分析。研究人员进行了敏感性分析以解决多效性问题。

吸烟与总体乳腺癌风险的关联

研究人员发现遗传预测的LSI与乳腺癌风险增加相关(每增加一个SD的OR为1.18，95%CI为1.07-1.30，P=0.11×10-2)，但没有证据表明遗传预测的CPD与乳腺癌风险增加相关(OR为1.02, 95%CI为0.78-1.19, P=0.85)。敏感性分析得出了类似的结果，并没有强有力的证据表明存在多效性效应。

由此可见，该MR研究提供了支持证据，表明吸烟者终生吸烟与乳腺癌风险之间存在潜在的因果关系，但每天吸烟量与其无关。
