OMCL:代谢综合征妇女血清胎球蛋白-B水平升高并与氧化应激增加相关

背景:以往对血清胎球蛋白-B(胎球蛋白样蛋白IRL685)与T2 DM的关系已有研究,但血清胎球蛋白-B变化的原因及其在代谢性疾病中的调节因子尚不清楚。

目的:在这里,我们评估了新诊断的甲胎蛋白综合征患者的血清胎球蛋白B水平,并进行了多种干预,以探讨胎球蛋白-B在甲胎蛋白综合征发病机制中的作用。

方法:采用酶联免疫吸附试验(ELISA)测定血清胎球蛋白-B(F-B)水平。采用生物信息学方法分析胎球蛋白B相关基因和信号转导途径。此外,在体外实验中还测量了氧化应激参数。对于亚组,我们进行了EHC、OGTT和GLP-1RA治疗,以调查血清胎球蛋白-B的调节因素。

结果:我们发现,与健康人相比,代谢综合征妇女血清胎球蛋白-B水平显著升高。此外,血清胎球蛋白-B与腰臀比、脂肪百分比、甘油三酯、空腹血糖、糖化血红蛋白、FINS、HOMA-IR、VAI和LAP呈正相关。生物信息学分析表明,大多数胎球蛋白B相关核心基因参与胆固醇代谢和脂肪分解。与这一发现一致的是,多元回归分析显示,甘油三酯含量和腰臀比与血清胎球蛋白-B独立相关。我们还观察到健康受试者在葡萄糖负荷后血清胎球蛋白-B水平显著升高,在体外,胎球蛋白-B过表达促进了HepG2细胞的氧化应激。在使用GLP-1RA治疗6个月后,代谢综合征女性患者的血清胎球蛋白-B水平随着代谢和胰岛素敏感性的改善而降低。

结论:因此,血清胎球蛋白-B与蛋氨酸有关,蛋氨酸可作为氧化应激的生物标志物。
